The Dutch winger has suffered an injury to his leg and will require the best part of a month to recover from the knock.
The 22-year-old was an unused substitute for Real's 1-0 defeat to Lyon in the UEFA Champions League on Tuesday, but has been unable to train since returning to Spain.
He underwent medical tests on Friday in an effort to get to the bottom of the problem and has been informed that he is carrying a muscle complaint.
The injury to his left leg will keep him out of action for the rest of February at least, preventing him from staking a claim for a regular role at Real.
Drenthe will miss Primera Liga games against Villarreal and Tenerife but could be back in time for the return date with Lyon on 10th March.
